In this webinar, Peter Favreau of ZEISS and Olivier Frey of InSphero examine how standardized 3D cancer models, fast volumetric microscopy, and advanced image analysis can be combined to support more efficient and quantitatively robust tumor microtissue screening.
Three-dimensional tumor systems can reveal treatment effects that may be missed in conventional monolayer cultures. However, applying these models across large screening studies introduces a practical challenge: traditional confocal microscopy records multiple focal planes sequentially, making complete volume acquisition time-intensive.
The speakers present a 384-well drug-screening study using reproducible colorectal tumor and fibroblast co-cultures developed with InSphero technology. Samples were recorded using the ZEISS LSM Lightfield 4D, allowing the full depth of each tumor microtissue to be acquired without serial z-plane imaging, before three-dimensional analysis in ZEISS arivis Pro.
The resulting workflow enables concentration-dependent and tumor-selective treatment effects to be measured across an entire screening plate. The webinar also demonstrates how reducing dense tumor microtissues to two-dimensional projections can obscure overlapping cells, introduce errors in cell counts, and affect the interpretation of quantitative results.
